You want to perform a lookup of all contacts for whom a field does not contain specific data. For example, you want to find all contacts who do not have "Supervisor" in the Title field.
To do this, you will need to create a SmartQuery. Please follow these steps:

Choose the Lookup menu, and choose Other.
In the field in which you want to search, enter the text that you want to exclude from the lookup, for example, Supervisor.
Choose the Query menu, and choose Convert To SmartQuery. A new window and the SmartQuery Assistant will appear, with a basic query already entered in the window. By default, this will be an "equals" query. For example, if you entered Supervisor in the Title field, the query would be: (Title = Supervisor).
Select the equals (=) operator. In the Logical Operator box of the SmartQuery Assistant, select <> and click Insert.
Save or run the query:

To save the query for use later, choose the File menu, and choose Save As. Enter a File Name and click Save.
To run the query, click Execute.
